sheesh, i don't know how i could pick just one... how about by genres?
comedy: Raising Arizona (this movie just slays me every time i watch it)
drama: Godfather I & II (III was an abomination, IMO )
romance: Sleepless in Seattle (i'm a sucker for that whole mushy thing)
western: Unforgiven (i'd watch Clint Eastwood read a cereal box)
horror: Alien (still gives me the heebie jeebies all these years later)
