Do you mean SIDE buttons? If so, then yes you can. Assuming you're using WMP, go into Options and look for the Buttons tab. You can map the volume to the side buttons. Most multimedia apps also have a button mapping feature.
I've tried that already. It does not work. It does not override the system definition. It calls up the Wireless Enable/Disable & Notes.
